2022年08月受聘于中国海洋大学“青年英才工程”第一层次教授岗位,加入医药学院开展独立研究。主要研究方向为血栓性心脑血管病发病机制,以及靶向细胞黏附受体的抗心脑血管病海洋生物新药研发。本研究团队具备完整的,从相关病理机制探索,到靶向药物分子发掘与设计,再到基于细胞和动物模型的药理及毒理学评价技术体系。近年来发表SCI论文26篇,其中以第一(含共一)/通讯作者在Blood, Nature immunology, Circulation Research, EBioMedicine, Thrombosis and Haemostasis, Cellular and Molecular Life Sciences, Archives of Toxicology, Toxins, Chinese Journal of Natural Medicines等顶尖和权威期刊发表研究论文。在新型抗血小板血栓先导化合物发掘与设计方向获授权国家发明专利3项。 | |||||||

血栓是高危心脑血管疾病(如心梗、脑梗)的一个主要病因。血小板在血栓形成尤其是动脉栓塞发生中不可或缺。αIIbβ3整合素是血小板及其前体细胞中特有的黏附受体。我们首次发现β3整合素、14-3-3ζ蛋白以及c-Src激酶形成复合体并在血小板激活和血栓形成中发挥关键作用。14-3-3ζ蛋白通过同时结合于β3整合素胞内“TST”结构域和c-Src的SH2结构域促进14-3-3ζ-c-Src-integrin-β3复合体的形成以及αIIbβ3整合素外向内的信号传导。针对此复合物设计发掘的抑制剂可以显著抑制血小板聚集、延展和血栓的形成,同时不会明显影响αIIbβ3与其胞外配体的结合。此研究为开发新型、低出血风险的抗血栓药物提供了新靶点和思路,同时也提供了一系列潜在的抗血小板/抗血栓先导分子。(相关一系列研究成果发表在Blood 2020, Thrombosis and Haemostasis 2020, Cardiovascular, Haematological Disorders-Drug Targets2020,Molecules 2021,并已被授权3项发明专利201710070868.2,202010805688.6,202010805686.7)。
基于天然生物药材的药用功能和生物的生存策略,从海洋生物及动物毒液/分泌物中定向发掘活性分子,并研究它们对血液和心脑血管系统的药理、毒理学作用。基于此技术体系分别筛选到了一系列具有抗血栓或促凝血等功能的生物分子。发现血小板膜糖蛋白激活和去唾液酸糖基化是部分毒素引起血栓性微血管病和血小板减少的新机制。为血栓性心脑血管病的防治提供了一系列的先导药物分子及模板,并为食/药用动植物资源活性成分开发和新型靶向药物的高效发掘提供了理论和技术支持。(相关一系列研究成果发表在Cellular and Molecular Life Sciences 2022,Blood 2021, Archives of Toxicology 2021,Scientific Reports 2021,Thrombosis and Haemostasis 2020,Toxins 2020,The international journal of biochemistry & cell biology 2014等)。 | |||||||
